Expertise: Biogenesis of proteins of the mitochondrial inner membrane, membrane integration and assembly of mitochondrial translation products. Biogenesis of proteins of the intermembrane space of mitochondria, characterization of the machinery which catalyses protein oxidation and analysis of its relevance in vitro and in vivo.
|Methodology provided: The Herrmann lab combines yeast genetics and protein biochemistry to study fundamental questions of molecular cell biology. Methods used in the group include: purification of mitochondria; systematic or random genetic screens in yeast; purification of protein complexes by native gel electrophoresis, coimmuno precipitation or affinity purification; purification of ribosomes on sucrose gradients; the analysis of protein translation in mitochondria and yeast cells; in vitro transcription/translation of proteins; import assays with purified mitochondria; enzyme assays to measure a variety of different metabolic activities; expression and purification of proteins from bacteria and fungi; measuring protein folding by light scattering and solubility assays; monitoring thiol oxidation in proteins in vitro and in vivo; trapping of sulfenylated proteins in vivo and in cellular extracts by use of protein traps of chemicals; SILAC labeling of yeast proteins and mass spectrometry-based proteomics.|
