Schwappach, Blanche Göttingen

Expertise: Membrane protein biogenesis, particularly tail-anchored proteins; sorting of membrane proteins in the secretory pathway; cellular ion and redox homeostasis, model organisms: yeast and mouse

Methodology provided: The Schwappach lab will contribute yeast synthetic genetic analysis (SGA) technology and an automated-microscopy screening platform to the consortium. Yeast strains of interest, i.e. redox-relevant deletion mutants, strains expressing GFP fusions or sensor constructs, can be systematically crossed into libraries of deletion mutants or strains carrying hypomorphic alleles or GFP fusions to ORFs. The resulting libraries of interest can then be automatically screened under different conditions to uncover genes or pathways relevant to redox homeostasis and regulation. More generally, we routinely prepare membranes by subcellular fractionation and can help to optimize the solubilization, transfer and detection of membrane proteins or their complexes using SDS PAGE or Blue Native PAGE gel electrophoresis.
